Muco-adhesive buccal tablets of candesartan cilexetil for oral delivery: preparation, in-vitro and ex-vivo evaluation

چکیده
Candesartan cilexetil (CC) is an angiotensin II-receptor blocker (ARB). The antihypertensive effect of CC 4-16 mg/day was as great that other once-daily dosage regimens. has high first-pass metabolism and low oral bioavailability. bioavailability such drugs may be significantly improved if delivered through the buccal route; hence mucosal delivery one alternative methods systemic drug delivery. This study’s objective to develop mucoadhesive tablets candesartan using carbopol-934P, hydroxyl propyl methyl cellulose (HPMC), Eudragit RLPO, sodium carboxy (Na-CMC) polymers. Prepared tablet formulations were evaluated for optimized system based on physicochemical properties, ex-vivo residence time, in-vitro, ex vivo permeation studies. evaluation parameters within acceptable Pharmacopoeial limits. However, swelling bio-adhesive time increased with increasing polymer concentrations. in-vitro release research shown exhibited a higher than all have been considered formulation. mechanism from kinetic suggests follows zero-order kinetics diffusion mechanism. Further, in-vivo in animal fashions required prove performance formulation.
